摘要
利用单因素筛选和正交试验,对球形芽孢杆菌(Bacillus sphaericus)NBIM-311高毒菌株的发酵培养基进行优化,得到其最佳培养基配方为鱼粉20.0 g/L,玉米浆15.0 g/L,花生粕60.0 g/L,酵母膏7.5 g/L,碳酸钙1.0 g/L,硫酸镁0.3 g/L,磷酸二氢钾0.1 g/L。通过200 L和5 000 L发酵罐分别发酵,发酵液对致倦库蚊致死中浓度分别为0.963 mg/L和0.852 mg/L;发酵液经过后处理所制备的原粉对致倦库蚊致死中浓度分别为0.018 9 mg/L和0.022 3 mg/L。
The fermentation medium for highly toxic Bacillus sphaericus strain NBIM-311against Culex quinquefasciatus was optimized through single factor screening and orthogonal experiments.The optimum medium was composed of20.0g/L fish m eal,15.0g/L com steep liquor,60.0g/L peanut m eal,7.5g/L yeast extract,1.0g/L Calcium carbonate,0.3g/L Magnesium sulphate and0.1g/L Potassium dihydrophosphate.The pilot fermentation of the strain was successfully carried out in200L and5000L ferm entor,the LCf,〇of the fermentation broth were0.963m g/L and0.852m g/L respectively.The LCf,〇of the prepared raw powder obtained from post-treatm ent of the fermentation broth were0.0189m g/L and0.0223m g/L respectively.
