棘孢木霉发酵培养基的响应面法优化

摘要 为了得到棘孢木霉(Trichoderma asperellum)发酵的最佳培养基,应用Plackett-Burman设计法对影响棘孢木霉菌丝体产量的关键因子进行了筛选,并进一步采用响应面分析法对影响菌丝产量的关键因素最佳水平进行了研究。结果表明,影响棘孢木霉菌丝体产量的关键因素为硝酸铵和硫酸镁的浓度。通过响应面分析法的拟合和推算,得到在硝酸铵和硫酸镁浓度分别为1.7 g/L和1.43 g/L时,模型预测发酵最佳产量为0.85 g/mL,验证值为0.905 g/mL,预测值与验证值之间吻合较好,约是原始培养基产量的2倍。 In order to obtain the optimal medium composition for T rieho derm a a sp erellu m production,the Plackett-Bunnan design was used to evaluate the effects of seven variables,and the optimum levels of key factors affecting mycelium yield were conducted by response surface analysis methodology.The results showed that,the optimal values of the two parameters determined by response surface analysis methodology were ammonium nitrate1.7g/L and magnesium sulfate1.43g/L.The mycelium production was0.85g/mL,and the actual experimental result was0.905g/mL,they were in agreement and it is about two folds of the production before optimization.
