摘要
目的探讨独正杆不同极性部位促进SD大鼠骨折愈合活性。方法以SD大鼠为研究对象,制作左前肢桡骨骨折模型,通过血清钙水平及碱性磷酸酶(AKP)活性测定、骨折组织HE染色、半定量RT-PCR测定VEGF mRNA表达来探讨独正杆不同极性部位灌胃给药对SD大鼠骨折愈合的影响。结果与对照组比较,独正杆正丁醇部位及乙酸乙酯部位可以显著升高AKP血清浓度(P<0.05),提前钙浓度峰值发生时间及钙浓度降低时间(P<0.05),提前软骨细胞及纤维性骨痂发生时间,增加骨痂生长速度、提前骨折部位塑形及骨髓腔贯通时间,上调VEGF mRNA表达水平(P<0.05)。结论独正杆正丁醇部位及乙酸乙酯部位对SD大鼠桡骨骨折愈合有显著促进作用。
Objective To investigate the different polar parts extracted from radix Vitis davidii Foёx on fracture healing in SD rats.Methods A model of the radial fracture of the left forelimb was made in SD rats.The activity of different polar parts extracted from radix Vitis davidii Foёx on fracture healing in fracture SD rats was research by the determination of the Calcium ions and Alkaline phosphatase concentration in serum,the HE staining of fracture tissue,and RT-PCR detection was used to detect VEGF mRNA expression of radius fracture organization.Results Compared with the blank group,the serum concentration of AKP was significantly increased by n-butanol extract and ethyl acetate extract of radix Vitis davidii Foёx(P<0.05),the time of peak calcium concentration and the decrease of calcium concentration were advanced(P<0.05).The peak time of calcium concentration and reduced time of calcium concentration the was improved(P<0.05),the occurrence time of cartilage cells and fibrous callus were increased,the growth rate of callus were increased,the shaping time of fracture site and the time of bone marrow cavity perforation were advanced,and the expression level of VEGF mRNA was increased(P<0.05).Conclusion Ethyl acetate extraction and n-butanol extraction from radix Vitis davidii Foёx have a significant effect on the healing of radius fracture in SD rats.
