牛至草醇提物的抗氧化与抗炎、止咳祛痰作用研究 被引量:1

摘要 目的:研究牛至草醇提物(NZEt)的体外抗氧化及抗炎、止咳祛痰作用。方法:用95%乙醇制备NZEt;采用DPPH·、羟基自由基(·OH)清除能力、还原能力检测方法考察NZEt的抗氧化作用,并对其总黄酮、总多酚含量进行测定;通过二甲苯致小鼠耳廓肿胀试验、氨水致咳试验及气管酚红排泌试验,探讨其抗炎、止咳、祛痰作用。结果:NZEt的DPPH·、·OH清除能力和铁离子还原能力均随浓度增加呈逐渐增高趋势。NZET最高浓度(2mg/ml)时清除DPPH·自由基能力接近于相同浓度的VC清除DPPH自由基能力,而其清除·OH能力、还原能力均小于浓度为0.1mg/ml VC的清除OH·能力和铁离子还原能力。NZEt中总黄酮和多酚含量分别为46.92mg/g、32.64mg/g。NZEt能够抑制二甲苯致小鼠的耳廓肿胀度(P<0.05)、减少氨水喷雾致小鼠咳嗽次数(P<0.05)、促进小鼠气管酚红排泌(P<0.05)。高、中剂量NZEt止咳、祛痰作用优于阳性对照药急支糖浆(P<0.05)。结论:NZEt具有较显著的体外抗氧化活性与抗炎、止咳、祛痰的作用。 Objective:To study the antioxidant and antiinflammatory,antitussive and phlegm effects of ethanol extract of Nile.Methods:NZEt was prepared with95%ethanol.The antioxidant effects of NZEt were studied by DPPH·hydroxyl radical,reducing ability and detection method.The total flavonoids and total polyphenols were determined.Swelling test,ammonia induced cough test and bronchial phenol red excretion test,to explore its anti inflammatory,cough,expectorant effect.Results:NZEt had a clear activity against three free radicals,and all of them increased gradually with the increase of concentration.The highest concentration(2mg/ml)cleared DPPH·The ability to close to the same concentration of VC,and its removal of the OH ability,the reduction capacity was less than the same concentration of VC.The total flavonoids and polyphenol contents of NZEt were46.92mg/g and32.64mg/g,respectively.it can inhibit the swelling of ear(P<0.05)and reduce the number of cough caused by ammonia spray(P<0.05),and promote the phenol red excretion(P<0.05).High and medium dose NZEt cough,expectorant effect is better than the positive control drug acute syrup(P<0.05).Conclusion:NZEt has more significant antioxidant activity in vitro and anti inflammatory,cough and expectorant.
