复方斑蝥胶囊对原发性肝癌患者介入治疗后细胞免疫功能的影响

The influence on cellular immune function of Fufang Banmao capsules in hepatocellular carcinoma patients with interventional therapy

目的:观察原发性肝癌(HCC)患者介入治疗及联合复方斑蝥胶囊治疗前后细胞免疫功能的变化。方法:本研究纳入20例健康志愿者及42例原发性肝癌患者,肝癌患者随机分为两组,分别为介入治疗组(21例)和介入治疗后联合复方斑蝥胶囊治疗组(21例),复方斑蝥胶囊用量为1g/次,3次/天,连续服用4周。在治疗前后分别收集外周血,分离外周血单个核细胞(PBMC),应用流式细胞术检测外周血CD_3^+T细胞、CD4+T细胞、CD_8^+T细胞、NK细胞、NKT细胞和B细胞的比例。结果:两组患者治疗前后肝脏功能(包括ALT、AST和T-BIL)无明显变化。原发性肝癌患者外周血CD_8^+T细胞、NK细胞和B细胞比例较健康志愿者显著降低,接受介入治疗的患者治疗后外周血免疫细胞的比例与治疗前比较无明显变化,而接受介入联合复方斑蝥胶囊治疗的患者外周血CD_8^+T细胞(31.8±7.78)%vs(25.0±4.45)%,P<0.05)和NK细胞(8.87±3.61)%vs(5.32±0.88)%,(P<0.05)比例较治疗前显著升高。结论:HCC患者存在细胞免疫功能障碍,介入治疗并不能恢复HCC患者受抑制的细胞免疫功能,而复方斑蝥胶囊可部分恢复介入治疗后HCC患者的细胞免疫功能。

Objective:To investigate the changes in cellular immune function in hepatocellular carcinoma(HCC)patients with interventional therapy in combination with Fufang Banmao capsules therapy.Methods:A total of twenty healthy individuals and forty two HCC patients were enrolled in this study.HCC patients were divided into two treatment groups.One was interventional therapy group(n=21),and the other was interventional therapy in combination with Fufang Banmao capsules therapy group.The usage of Fufang Banmao capsules was1g per time and three times daily for a total of four weeks.Peripheral bloods were collected before and after therapy,and peripheral blood mononuclear cells(PBMC)were isolated.The percentages of CD3+T cells,CD4+T cells,CD8+T cells,NK cells,NKT cells,and B cells were measured by flow cytometry.ResultsThe liver function(including ALT,AST,and T BIL)did not change significantly before and after therapy in HCC patients.The percentages of peripheral CD8+T cells,NK cells,and B cells were notably reduced in patients with HCC in comparison with those in healthy individuals.There were no remarkable differences of peripheral immune cells before and after interventional therapy in HCC patients.Moreover,the percentages of CD8+T cells(31.8±7.78)%vs(25.0±4.45)%,(P<0.05)and NK cell(8.87±3.61)%vs(5.32±0.88)%,(P<0.05)were significantly elevated in HCC patients who received interventional therapy in combination with Fufang Banmao capsules therapy.Conclusion There was cellular immune dysfunction in HCC patients.Interventional therapy could not restore the inhibited cellular immune function of HCC patients.Fufang Banmao capsules partly recovered the cellular immune function in HCC patients received interventional therapy.
