摘要
目的分析子宫内膜癌(EC)患者合并与未合并代谢综合征(MS)的内膜组织miRNA表达谱差异,为研究MS相关EC疾病发生机制提供依据。方法收集合并MS的EC和未合并MS的EC病灶内膜各3例,1例正常内膜组织对照。提取总RNA,利用miRNA基因芯片检测miRNA表达谱差异。结果两组患者内膜组织与正常内膜组织间均有差异表达的miRNA,其中合并MS者与正常内膜组织间存在51个miRNA差异表达,其中15个表达上调,36个表达下调。未合并MS者与正常内膜组织间存在9个miRNA差异表达,其中3个表达上调,6个表达下调。两种患者子宫内膜癌组织间存在10个miRNA差异表达,其中4个表达上调,6个表达下调。结论合并MS与未合并MS的EC组织间miRNA的表达差异可能与MS相关的EC发病机制相关,两组患者间差异表达的miRNA可为进一步研究子宫内膜癌发生的分子机制提供依据。
Objective To study miRNA expression patterns in endometrial carcinoma(EC)combined with the Metabolic Syndrome(MS)and noncombined with MS in order to explore the role of the microRNAs in EC combined with the MS.Methods Three freshfrozen samples of endometrial carcinoma Combined with MS and3cases of nonCombined with MS and1case of normal endometrium were collected.Total RNA was extracted from samples and miRNA microarray was used to evaluate the difference of expression spectrum of miRNA.Results There were different expression of miRNA among the two groups of endometrium and normal endometrium.There were51miRNA different expressions between EC combined with the MS and the normal endometrium,and among them there are15upregulated miRNA and36downregulated miRNA.There were9miRNA different expressions between EC noncombined with the MS and the normal endometrium,and there are3upregulated miRNA and6downregulated miRNA among them.10different miRNA were revealed between the two type of EC,and there are4upregulated miRNA and6downregulated miRNA among them.Conclusion Different expressions of miRNA between EC Combined and nonCombined with MS may provide insights into their roles in the progression of EC.The study of miRNA contributes to elucidate the molecular mechanism of endometrial carcinoma.
