肺腺癌转移相关转录本1在乳腺癌患者血浆中的表达及临床意义 被引量:14

摘要 目的:探讨长链非编码RNA肺腺癌转移相关转录本1(metastasis-associated lung adenocarcinoma transcript1,MALAT-1)在乳腺癌患者血浆中的表达及其临床意义。方法:采用实时荧光定量PCR(qRT-PCR)方法检测60例乳腺癌患者癌组织及癌旁组织中MALAT-1的表达,60例乳腺癌患者及60例乳腺纤维瘤患者血浆中MALAT-1的表达;分析乳腺癌患者血浆MALAT-1表达量与临床病理特征的关系;采用电化学发光免疫测定法检测血浆中CA125和CA153含量,建立多元逻辑回归模型分析血浆中MALAT-1、CA125和CA153对乳腺癌的诊断效能及3项指标联合诊断价值。结果:乳腺癌患者癌组织MALAT-1表达量明显高于癌旁组织;与乳腺纤维瘤患者相比,乳腺癌患者血浆中MALAT-1表达明显增高,且血浆中MALAT-1表达与基因分型和淋巴结转移明显相关(P<0.05)。血浆MALAT-1单独诊断乳腺癌的ROC曲线下面积(AUC)为0.961(P<0.001),敏感度和特异性分别为96.7%和93.3%,其诊断价值高于CA125(AUC=0.618,P=0.022)和CA153(AUC=0.623,P=0.016);3项指标联合诊断敏感度(98.3%)高于单独检测。结论:乳腺癌患者血浆中MALAT-1呈高表达,其可能为乳腺癌诊断的潜在生物学标志物。 To investigate the expression levels of long non-coding RNA metastasis-associated lung adenocarcinoma transcript1(MALAT-1)in the plasma of patients with breast cancer and its clinical significance.Methods:MALAT-1levels in the60pairs of human breast cancer tissue and matched para-carcinoma tissues,in the plasma of60breast cancer patients and60plasma samples from the patients with breast fibroadenoma were detected by using quantitative real-time PCR.The correlations of plasma MALAT-1level with the clinicopathological features of the breast cancer patients were analyzed.The content of CA125and CA153in plasma was detected by electrochemiluminescence;and a multivariate Logistic regression model was established to analyze the diagnostic efficacy of plasma MALAT-1,CA125,CA153and the diagnostic value of three indicators combined.Results:MALAT-1expression in breast cancer tissue was higher than that in adjacent breast tissue.Compared with the breast fibroadenoma group,breast cancer patients had increased expressions of MALAT-1in plasma,and plasma MALAT-1level was significantly correlated with triple-negative breast cancer and lymph node metastasis(P<0.05).ROC curve and the multivariable Logistic regression model showed that the area under ROC curve(AUC)of plasma MALAT-1was0.961(P<0.001)for breast cancer diagnosis,with a diagnostic sensitivity and a specificity of96.7%and93.3%,respectively.The diagnostic power and specificity of plasma MALAT-1was much higher than those of CA125(AUC=0.618,P=0.022)and CA153(AUC=0.623,P=0.016);and the combination of the3indicators had higher sensitivity(98.3%)than the single indicator.Conclusion:MALAT-1was highly expressed in the plasma of breast cancer patients,and may serve as a potential biomarker for breast cancer diagnosis.
