摘要 为探究T-2毒素对凡纳滨对虾DNA黏度的损伤效应,通过小牛胸腺DNA与T-2毒素作用,采用响应面分析T-2毒素与DNA的最佳作用条件,通过体外T-2毒素与对虾DNA作用验证,以及喂食对虾T-2毒素进行蓄积染毒后,观察其对DNA黏度的影响。结果:响应面分析优化出对DNA黏度影响最明显的反应条件为T-2毒素质量浓度2.70 ng/mL、DNA质量浓度50μg/mL、反应时间43 min;T-2毒素与对虾DNA体外作用,随着T-2毒素质量浓度的增加,DNA黏度随之增大,在T-2毒素质量浓度为2.70 ng/mL时达到最大值,此后降低;喂食对虾T-2毒素进行蓄积染毒对对虾DNA黏度的影响无明确规律,与体外对虾DNA实验结果并不相似。本研究阐明T-2毒素对对虾DNA黏度的影响,对探讨T-2毒素的毒理作用具有一定的意义。 The objective of this paper was to explore the effect of T-2toxin on the viscosity of DNA from shrimps(Litopenaeus vannamei).The conditions for the reaction between calf thymus DNA and T-2toxin were optimized using response surface methodology.The reaction between shrimp DNA and T-2toxin in vitro was investigated.Furthermore,the effect of T-2toxin on DNA viscosity was examined after feeding shrimps with T-2toxin for20days to cause accumulative poisoning.The results obtained showed that a T-2toxin concentration of2.70ng/mL,a DNA concentration of50μg/mL and a reaction time of43min were determined to be the optimum conditions for the greatest effect of T-2toxin on DNA viscosity.DNA viscosity rose with increasing concentration of T-2toxin,reaching its maximum level at2.70ng/mL,followed by a decrease.The effect of T-2toxin accumulation in shrimps on the viscosity of DNA was not clear.This result was dissimilar to the result in vitro.In conclusion,this study may be of great significance for understanding the toxicological effect of T-2toxin on shrimps.
