摘要
目的:分析丹玄口康(DXKK)对ANE诱导的OSF大鼠的影响。方法:选择72只大鼠,随机分为正常组8只、药物治疗组32只和实验对照组32只,药物治疗组和实验对照组再分1、2、4、8周4个时间点组,每组8只;正常组不作任何干预,1周后取材,其余组大鼠乙醚麻醉后,用蘸有ANE的小刷子获取颊黏膜组织样本;药物治疗组中以DXKK 8g/kg给药,实验对照组以等量的蒸馏水灌胃,每天1次至时间点;检测不同组大鼠开口度,进行颊黏膜检测评分、处理颊黏膜组织后,经Masson三色法检测胶原蛋白、ELISA法检测羟脯氨酸,详细观察并记录检验结果。结果:(1)两组大鼠开口度逐渐缩减,其中实验对照组于第4周开始开口度小于正常组(P<0.05);随着ANE刺激时间的延长,开口度呈递减状,至第8周约16.3mm,开口度明显小于正常组(P<0.05);第3周起,实验对照组大鼠颊黏膜评分与正常组相比差异明显(P<0.05);实验对照组大鼠不同时间点检测Masson染色蓝色区域相对面积逐渐上升;从第2周开始实验对照组羟脯氨酸含量明显增大,与正常组相比差异有显著性意义(P<0.05)。结论:ANE刺激是大鼠形成OSF的重要原因之一;DXKK可通过抑制胶原的合成降低OSF的发生率。
Objective:To investigate the effect of the Danxuan Koukang pill on ANE-induced OSF in rats.Methods:72rats were randomly divided into the normal group(n=8),treatment group(n=32)and experimental control group(n=32).The experimental control group and the treatment group were divided into4groups,8cases in each group.The rats in the treatment group were treated with DXKK8g/kg.The rats in the experimental control group were treated with the same amount of distilled water.The opening degree of the rats in different treatment groups was measured.By the buccal mucosal examination,the buccal mucosa was treated and the collagen was detected by Masson three-color method.Hydroxyproline was detected by ELISA.Results:(1)The opening degree in the treatment groups was gradually reduced,and that in the experimental control group was smaller than that in the normal group at4th week(P<0.05).With the prolongation of the ANE stimulation time,the opening degree was decreased,and was16.3mm at8th week,smaller than the normal group(P<0.05).Inthe experimental control group,the buccal viscosity score was significantly different from the normal group(P<0.05).The relative area of Masson-stained blue region was gradually increased.The content of hydroxyproline in the experimental control group was significantly increased from the second week,with a difference between the experimental control group and normal group(P<0.05).Conclusion:ANE stimulation is one of the important reasons for the formation of OSF in rats.DXKK can reduce OSF by inhibiting the synthesis of collagen.
