摘要
目的研究半仿生-酶法提取虎杖中大黄素的最佳工艺。方法以HPLC法检测大黄素含量为指标,优选出纤维素酶用于半仿生-酶法中,采用正交实验对半仿生-酶法提取条件进行优化,并与超声提取法、微波提取法和醇提法进行比较。结果半仿生-酶法较其他提取方法效率更高,其最优条件为:提取温度70℃,料液比1∶18,提取时间2h。结论半仿生-酶法提取虎杖中大黄素的方法经济、有效、提取效率高,可作为大黄素的提取工艺应用。
Objective To study the suitable semi-bionic enzyme extraction method of emodin from Reynoutria japonica Houtt..Methods The content of the emodin in all the extracts was determined by HPLC.The cellulase was selected and the semi-bionic cellulase extractions were optimized by orthogonal experiment.Then the semi-bionic enzyme extraction method was compared with ultrasonic extraction,microwave extraction and ethanol extraction methods.Results The method of semi-bionic enzyme extraction is more efficient than the others.The preferred process of the orthogonal experiment was determined as follows:extraction temperature at70℃,solid-liquid ratio of1∶18and extraction time for2h.Conclusion Semi-bionic enzyme extraction is an economical and effective method for emodin,which can be used in the extraction process of Reynoutria japonica Houtt..
