化纤胶囊对脓毒症大鼠血小板最大聚集率、血小板活化复合物(PAC-1)、血小板表面蛋白CD62p的影响 被引量:3

摘要 目的 探究化纤胶囊对脓毒血症模型大鼠血小板最大聚集率、血小板活化复合物(PAC-1)、血小板表面蛋白CD62p的影响,阐明化纤胶囊对脓毒症大鼠血小板活化功能的调控机制。方法将140只实验动物SD大鼠随机均匀分成模型对照组、生理盐水干预组、血必净干预组、低剂量化纤胶囊干预组(30mg/kg)、中剂量化纤胶囊干预组(60mg/kg)、高剂量化纤胶囊干预组(90mg/kg)。CLP术后2h对药物干预组分别予相应药物进行灌肠,CLP术后12h后给予第2剂,连续3d,2次/d。模型制备后6h、12h、24h各时间点处死动物,取血进行血小板最大聚集率、血小板活化复合物(PAC-1)、血小板表面蛋白CD62p的检测。结果与模型组比较,血必净干预组与化纤胶囊(低、中、高)剂量干预组均对脓毒血症模型大鼠血小板最大聚集率、血小板活化复合物(PAC-1)、血小板表面蛋白CD62p有降低作用(P<0.05),其中化纤胶囊(低、中、高)剂量干预组优于血必净干预组,而化纤胶囊高剂量组对于降低上述指标作用最明显(P<0.01)。生理盐水干预组对上述指标作用不明显(P>0.05)。结论化纤胶囊对于脓毒血症大鼠血小板最大聚集率、血小板活化复合物(PAC-1)、血小板表面蛋白CD62p的表达水平有降低作用,具有预防血栓形成的作用。 Objective Chemical fiber capsule on sepsis model rat platelet maximum aggregation rate,platelet activation complex(PAC),platelet surface proteins CD62p,clarify chemical fiber capsule on sepsis in the rat platelet activation function of the regulatory mechanism Methods Even140experimental animal SD rats were randomly divided into model control group,normal saline intervention group,the blood will net intervention group,low-dose chemical fiber capsule intervention group(30mg/kg),dose of chemical fiber capsule intervention group(60mg/kg),high dose of chemical fiber capsule intervention group(90mg/kg).2h after CLP to the drug intervention group respectively for the corresponding drugs enema,12h after CLP postoperative give agent2,3days in a row,twice a day.Model6h,12h,24h after preparation of each point in time to be put to death animals,blood platelet maximum aggregation rate,platelet activation complex(PAC),platelet surface proteins of CD62p detection.Results Net intervention group compared with model group,the blood will with chemical fiber capsule(low,middle and high dose of intervention group were on sepsis model rat platelet maximum aggregation rate,platelet activation complex(PAC),platelet CD62p surface proteins have lower(P<0.05),including chemical fiber capsule(low,middle and high dose of intervention group is better than that of blood will net intervention group,and chemical fiber capsule high-dose group for the most obvious effect of the above indexes(P<0.01).Physiological saline intervention group on the above index effect is not obvious(P>0.05).Conclusion Chemical fiber capsule for sepsis rats platelet maximum aggregation rate,platelet activation complex(PAC),platelet CD62p expression level had lower surface proteins,have the effect that prevent thrombosis.
