摘要
为了检验嗜酸红假单胞菌合成5-氨基乙酰丙酸(5-ALA)的能力,测定发酵液中5-ALA的含量,首先对5-ALA的检测方法进行了优化;其次对嗜酸红假单胞菌合成5-ALA的时间和产量进行了试验,并对5-ALA在其发酵液中的稳定性进行了研究。结果表明:在Ehrlich’s试剂比色法的基础上,采取70℃水浴显色5 min,便能较好地满足检测要求;5-ALA的最佳合成时期处于菌株的对数生长期,5-ALA的产量最高可达50.2 mg/L;发酵液pH值为4.2,室温避光保存可以使5-ALA在嗜酸红假单胞菌发酵液中稳定保存。
In order to test the ability of synthesizing5-ALA by Rhodopseudomonas acidophila,and determine the content of5-ALA in fermentation broth,we first optimized the test method of5-ALA;secondly,the time and yield of5-ALA synthesized from Rhodopseudomonas acidophila were tested,and the stability of5-ALA in the fermentation broth was studied.The results showed that,on the basis of Ehrlich's reagent colorimetric method,using70℃water bath to show color5min,it can meet the testing requirements well.The optimal period of synthesis of5-ALA was in the logarithmic growth period of the strain,and the highest yield of5-ALA could reach50.2mg/L.The pH value of the fermentation broth is4.2,and the room temperature preservation can keep5-ALA stable in the fermentation broth of Rhodopseudomonas acidophila.
