敦煌医方大补肾汤对镉染毒模型大鼠肝组织细胞因子和氧化应激的影响 被引量:3

The Influence of Dunhuang Medical Prescription DaBuShen Decoction on Oxidative Stress and Cytokines in Liver Tissue of Cadmium Poisoning Rat Model

摘要 目的:探讨大补肾汤对镉染毒模型大鼠肝组织细胞因子和氧化应激的影响。方法:将SPF级Wistar大鼠60只随机分为正常对照组、模型组、乙酰半胱氨酸组及大补肾汤高、中、低剂量组。末次给药24小时后处死大鼠,计算胸、脾指数。采用比色分析法测定大鼠肝组织超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量,酶联免疫吸附试验(ELI SA)测定大鼠肝组织白细胞介素2(IL-2)和转化生长因子β_1(TGF-β_1)的含量。结果:与正常对照组比较,模型组大鼠体质量明显减轻(P<0.05);大补肾汤中、高剂量组大鼠体质量明显高于模型组(P<0.05)。模型组胸、脾指数低于正常对照组,差异有统计学意义(P<0.05);大补肾汤各剂量组大鼠脾脏指数高于模型组,且高剂量组胸腺指数高于模型组,差异有统计学意义(P<0.05)。与正常对照组比较,模型组MDA含量升高、SOD活性降低(P<0.05);大补肾汤各剂量组MDA含量低于模型组,SOD活性高于模型组(P<0.05)。模型组IL-2含量低于正常对照组,TGF-β_1含量高于正常对照组(P<0.05);大补肾汤各剂量组IL-2含量高于模型组,TGF-β_1含量低于模型组(P<0.05)。结论:大补肾汤对镉所致大鼠的氧化损伤和免疫损伤具有保护作用。 Objective:To discuss the influence of DaBuShen decoction on oxidative stress and cytokines in liver tissue of the rat model suffering cadmium poisoning.Methods:Sixty Wistar rats were randomized into normal control group,the model group,acetylcysteine group,high,moderate and low doses groups of DaBuShen decoction.The rats were sacrificed in24hours after the last medication,spleen and thymus indexes were calculated,the activity of SOD and the contents of MDA in liver tissue of the rats were measured by using colorimetric analysis,the contents of IL-2and TGF-β1in liver tissue were detected by ELISA method.Results:Compared with normal control group,body weight of the rats in the model group reduced obviously(P<0.05);high and moderate dose groups of DaBuShen decoction were notably higher than the model group in body weight of the rats remarkably(P<0.05).The model group was lower than normal control group in spleen and thymus indexes obviously,and the difference showed statistical meaning(P<0.05);different doses groups of DaBuShen decoction were higher than the model group in the spleen index of the rats,and high dose group of DaBuShen decoction was higher than the model group in thymus index obviously,and the difference showed statistical meaning(P<0.05).Compared with normal control group,the contents of MDA lifted apparently while the activity of SOD lowered notably in the model group(P<0.05);different doses groups of DaBuShen decoction were lower than the model group in the contents of MDA notably,higher than the model group in the activity of SOD notably(P<0.05).The model group was lower than normal control group in the contents of IL-2notably,higher than normal control group in the contents of TGF-β1(P<0.05);different doses groups of DaBuShen decoction were higher than the model group in the contents of IL-2notably,lower than the model group in the contents of TGF-β1(P<0.05).Conclusion:DaBuShen decoction shows protective effects to oxidative damage and immunologic injury of the rat models suffering cadmiumpoisoning.
