凤丹愈伤组织中丹皮酚含量的测定 被引量:6

摘要 本文利用药用植物组织培养的方法,对凤丹子叶愈伤进行诱导,并用单因素实验和正交实验研究了愈伤组织中丹皮酚含量在不同水浴时间、超声时间及超声功率三个条件下的差异,并采用最优处理方式比较了丹皮酚在凤丹不同器官和子叶愈伤中的含量,以及不同激素配比和不同类型基本培养基对其含量的影响。结果表明:水浴2 h,超声30 min,100 W处理条件下丹皮酚含量最高,影响力大小为超声时间>水浴时间>超声功率,故该方法可用作愈伤组织中丹皮酚含量测定前样品的处理方法;丹皮酚含量在凤丹不同部位中差异很大,依次为:市售丹皮>叶片>种子>果荚>子叶愈伤;6-BA 0.5 mg/L+2,4-D 4.0 mg/L的激素配比和B5基本培养基不同程度上提高了子叶愈伤组织中丹皮酚的含量。 The callus of Paeonia ostii cotyledon was induced by tissue culturing of medicinal herbs.The content of paeonol in callus under different processing conditions,such as water bath time,ultrasonic time and ultrasonic power,was determined with single factor experiment and orthogonal experiment.Then,the optimal handling method was used to compare the content of paeonol in different organs and callus,and different effects on its content by growing on basic culture medium and different ratio of hormone.The results showed that the optimal processing conditions were water bath 2 h,ultrasonic treatment 30 min with ultrasonic power 100 W.The decreasing order of importance of the three processing conditions affecting content of paeonol was ultrasonic time,water bath time and ultrasonic power,successively.The above process can be used as a method of processing sample before detecting the content of paeonol in callus.The content of paeonol in different parts of P.ostii was quite different,and the decreasing order of content was Cortex Moutan,leaf,seed,pod and cotyledon callus,successively.The culture conditions of 6-BA 0.5 mg/L+2,4-D 4.0 mg/L and basic culture medium B 5 can improve the content of paeonol to varying degrees.
