HPLC法同时测定不同产地五味子中5种木脂素类成分的含量

Simuitaneous Determination of Five Lignans in Schisandra chinensis(Turcz.) Baill. from different Habitats by HPLC

目的:利用高效液相色谱法测定不同产地五味子中5种木脂素类成分的含量,并比较其差异。方法:采用菲罗门XB-C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈(A)-水(B),梯度洗脱(0~10 min,A为50%;10~15 min,A为50%~75%;15~25 min,A为75%;25~30 min,A为75%~50%);检测波长:217 nm;流速:1.0m L/min;柱温:30℃;进样体积:10μL。结果:5种木脂素类成分与相邻组分分离度均大于1.5,方法学考察结果均符合有关规定。结论:不同产地五味子中木脂素类成分含量存在较大程度的差异,建立的HPLC方法能够同时测定五味子药材中的5种成分的含量,可用于五味子药材及其制剂中木脂素类成分的含量测定和质量控制。

Objective To determine the contents of five lignans in Schisandra chinensis(Turcz.)Baill,and compare the differences among different habitats.Methods Phenomenex XB-C 18 column(4.6 mm×250 mm,5μm)was used.Acetonitrile(A)-water(B)was used as gradient mobile phase(0~10 min,50%A;10~15 min,50%~75%A;15~25 min,75%A;25~30 min,75%~50%A).The flow rate was 1.0 mL·min-1.The detection wavelength was 217 nm and the column temperature was 30℃.Results Chromatographic separation is greater than 1.5,the results of the methodology are consistent with the relevant regulations.Conclusion The HPLC method developed for simultaneous determination of 5 lignans is simple and valid.It can be used for quality evaluation and quality control of Schisandra chinensis(Turcz.)Baill,which can be used in the determination and quality control of the content of lignans in the medicine and its preparation.
