吸波面积法测定乌头的注射液及其颗粒中药效物质的含量 被引量:2

摘要 目的测定乌头的注射液及其颗粒中药效物质的含量,比较3种溶剂下吸波面积法所测含量的操作简便性与结果准确性。方法以水、甲醇和三氯甲烷为溶剂,获得2种剂型的药效物质,通过吸波面积法分别测定其含量,并对试验操作的简便性与结果的准确性进行比较。结果水、甲醇和三氯甲烷测得的吸波面积法线性方程分别为Y=0.321 1X+0.964 4(r=0.999 7),Y=0.432 7X+3.066 9(r=0.998 6),Y=0.145 4X+3.240 4(r=0.999 3);乌头注射液加样回收率分别为107.10%、112.51%、99.91%,乌头颗粒加样回收率分别为105.26%、106.61%、102.80%;乌头注射液中1 g生药材药效物质的含量分别为229.24、160.50、83.64 mg,颗粒的含量分别为222.66、141.19、74.85mg,注射液的药效物质含量比颗粒对应的药效物质含量大,随着溶剂极性的减小,含量的数值也减小,水为溶剂的操作最简便,含量最高,三氯甲烷的操作较为复杂,含量最低。结论开展的吸波面积法均结果准确、方法可行,代表了不同溶剂下获得的乌头药效物质整体含量的特征,所有药效物质的含量测定可使用水为溶剂,仅仅为乌头碱类生物碱的可选择三氯甲烷,而去除部分水溶性成分的则用甲醇为溶剂,为乌头类制剂不同极性所获得的各自药效物质的整体含量测定起到了较好的方法指导。 Objective To determine the content of medicinal substance in Aconitum injection and its granules,and compare the simplicity of operation and the accuracy of the results among the three kinds of solvents by area under the absorbance-wavelength curve(AUAWC).Methods Took water,methanol and chloroform as individual solvent,the medicinal substance in two dosage forms were obtained.The content of medicinal substances was measured by AUAWC,and the simplicity of operation and the accuracy of the results were compared.Results The linear equation of the AUAWC determined by water,methanol and trichloromethane were Y=0.321 1X+0.964 4(r=0.999 7),Y=0.432 7X+3.066 9(r=0.998 6)and Y=0.145 4X+3.240 4(r=0.999 3),respectively.The recoveries of Aconitum Injection were 107.10%,112.51%and 99.91%,respectively,and the recoveries of Aconitum Granules were 105.26%,106.61%and 102.80%.The contents of medicinal substance from 1 g raw aconitum herb of Aconitum Injection in three different solvent solution were 229.24,160.50,83.64 mg,respectively,and the contents of the granules were 222.66,141.19,74.85 mg,respectively.The content of medicinal substance in the injection was larger than that of the granules.As the polarity of the solvent decreased,the content of medicinal substance decreased.The operation with water was the easiest with the largest content,and the operation with chloroform was complex with the smallest content.Conclusion The method of AUAWC for three kinds of solvents was accurate and feasible.lt represented the characteristics of the total content of medicinal substances in aconitum obtained under different solvents.The deter^nination of the content of all medicinal substances can be used by water as the solvent,only aconitine alkaloid by the choice of chloroform,and the removal of part of the"water-soluble ingredients by methanol as the solvent.The study"would bring a good guidance for the determination of the total content of the respective medicinal substances from aconite preparations in different polarity parts.
