针刺捻转补泻法对自发性高血压大鼠小脑NE、5-HT及NO的影响 被引量:14

Effects of twirling reinforcing and reducingmethod of acupuncture on NE,5-HT and NO in cerebella of spontaneously hypertensive rats

摘要 目的观察针刺捻转补泻法对自发性高血压大鼠(spontaneously hypertensive rats,SHR)小脑去甲肾上腺素(norepinephrine,NE)、5-羟色胺(5-hydroxytryptaminw,5-HT)、一氧化氮(nitric oxide,NO)含量的影响,探讨小脑在针刺降压的效应机制。方法 40只9周龄雄性SHR随机分为模型组、针刺组、针刺补法组、针刺泻法组,每组10只;空白组WKY 10只。针前1天及针刺干预的第3、8、13、18、23、27天测量血压。28天后摘取小脑,应用Elisa法检测小脑组织NE、5-HT和NO的含量。结果 (1)血压:与正常组比较,模型组、针刺组、针刺补法组、针刺泻法组的收缩压均显著升高,差异有统计学意义(P<0.05);其中在针前,模型组、针刺组、针刺补法组、针刺泻法各组之间收缩压无统计学差异(P>0.05);自针后第3天开始至27天,针刺组、针刺补法组、针刺泻法组血压比模型组显著降低(P<0.05);自针刺后第8天开始至27天,针刺补法组、针刺泻法组与针刺组比较显著降低(P<0.05),而补法组与泻法组比较,未见统计学差异(P>0.05);自针后第13天开始至27天,针刺泻法组均比针刺补法组血压降低(P<0.05)。(2)NE、5-HT、NO:与空白组比较,其余各组小脑NE、5-HT含量显著升高(P<0.05),NO含量降低(P<0.05);与模型组比较,各针刺组NE、5-HT含量降低(P<0.05)NO含量升高(P<0.05);与针刺组比较,针刺泻法组NO含量升高(P<0.05);与针刺组和针刺补法组比较,泻法组小脑组织NE降低(P<0.05),而针刺组与补法组之间无统计学差异(P>0.05)。结论针刺捻转补法和捻转泻法均可有效降低SHR大鼠收缩压;自针后第7天开始,不同针刺手法降压效果出现显著差异,且捻转泻法组比捻转补法组降压效果显著,其降压作用可能与调节小脑,NE、5-HT、NO含量有关,且捻转补法组和泻法组的调节作用存在显著差异。 Objective To observe the effect of twirling reinforcing and reducing methods of acupuncture on norepinephrine(NE),serotonin(5-HT)and NO contents in the cerebella of spontaneously hypertensive rats(SHR);To discuss the mechanism of cerebellum in terms of reducing blood pressure through acupuncture.Methods Forty 9-week-old male SHR rats were randomly divided into 5 groups,namely normal group,control group,non-twirling group,twirling reinforcing method group,and twirling reducing method group,with 10 rats in each group.Blood pressure of the mice was measured 1 day before the treatment and on the 3rd,8th,13th,18th,23rd and 27th day after acupuncture intervention.28 days after the treatment,we took the cerebella of the rats and tested the NE,5-HT and NO content by Elisa.Results(1)Blood pressure:The systolic blood pressure of the rats in the control group,the non-twirling group,the twirling reinforcing method group and the twirling reducing method group was significantly higher than that of the normal group(P<0.05),which demonstrated statistical significance.There was no statistical difference between the four groups before the acupuncture.Between day 3 and day 27,the blood pressure of the rats in the non-twirling group,the twirling reinforcing method group and the twirling reducing method group was remarkably lower than that of the control group(P<0.05).Between day 8 and day 27,the blood pressure of the rats in the twirling reinforcing method group and the twirling reducing method group was significantly lower than that of the non-twirling group,while no statistical difference was shown between the twirling reinforcing method group and twirling reducing method group(P>0.05).Between day 13 and day 27,the blood pressure of the rats in twirling reducing method group was lower than that of the twirling reinforcing method group(P<0.05).(2)NE、5-HT、NO:Compared with the normal group,the NE and 5-HT content in the other four groups significantly increased(P<0.05)while NO content decreased(P<0.05);Compared with the control group,the NE and 5-HT content in the 3 groups that had taken acupuncture largely decreased(P<0.05)while NO content increased(P<0.05);Compared with the non-twirling group,the twirling reducing method group showed an increase in NO content(P<0.05);Compared with the non-twirling group and the twirling reinforcing method group,the twirling reducing method group presented a decrease in NE(P<0.05),while no statistical difference was shown between the non-twirling group and the twirling reinforcing method group(P>0.05).Conclusion Both twirling reinforcing and reducing method of acupuncture can effectively reduce the systolic blood pressure of SHR rats.Beginning from the 7th day after the treatment,huge difference appeared between rats receiving different acupuncture methods.Twirling reducing method is more effective at reducing the blood pressure than twirling reinforcing method and its antihypertensive mechanism may be related to the adjustment of NE,5-HT and NO content in cerebellum.There is marked difference between the adjusting function between twirling reinforcing method and twirling reducing method.
