鹰嘴豆多糖的分离纯化及其抗氧化作用研究 被引量:8

摘要 目的对鹰嘴豆果实多糖进行分离纯化得到均一多糖,对分离得到的均一多糖的抗氧化活性进行研究。方法利用热水对鹰嘴豆多糖进行提取,乙醇沉淀得到粗多糖;粗多糖经DEAE-Cellulose 52和Sephacryl S-300柱层析纯化,得到均一多糖CA-1b;高效凝胶渗透色谱法(HPGPC)验证CA-1b纯度和相对分子质量;酸水解、GC-MS对CA-1b进行糖组分分析;DPPH·清除评价均一多糖的体外抗氧化活性。结果鹰嘴豆果实中分离得到的多糖CA-1b为均一多糖,相对分子质量为1.33×106 Da,由阿拉伯糖(Ara)、甘露糖(Man)、葡萄糖(Glc)和半乳糖(Gal)组成,其摩尔比为2.2∶3.1∶6.2∶1.0。DPPH·清除实验表明,CA-1b具有良好的抗氧化活性,且呈现一定的量效关系,半数清除质量浓度IC50为18.2μg·mL-1。结论研究结果可为鹰嘴豆的深加工和进一步研究开发奠定理论基础。 To isolate and purify the homogeneous polysaccharides from Cicer arietinum and to evaluate the antioxidant activity.After the crude polysaccharides were obtained from the fruits of Cicer arietinum by water extraction,the polysaccharides were purified by DEAE-Cellulose 52 and Sephacryl S-300 to give the homogenous polysaccharide.High performance gel permeation chromatography(HPG PC)proved that the polysaccharides were homogenous.The carbohydrate content of polysaccharide was measured by phenol-sulfuric acid method.The monosaccharide composition of the homogenous polysaccharide was analyzed by GC and GC-MS after hydrolysis.The antioxidant activity of the homogenous polysaccharide was measured by DPPH.scavenging.The results showed that the polysaccharides(CA-lb)isolated from Cicer arietinum was homogeneous with relative molecular weight of 1.33X10 6Da.CA-lb was made up of Ara,Man,Glc and Gal.And their ratio was 2.2:3.1:6.2:1.0.DPPH.scavenging experiments showed that the CA-lb had a good antioxidant activity and there was a certain relationship between the activity and concentration with IC5018.2μg.mL 1.These results provide a theoretical basis for further research and development of Cicer arietinum.
