单硝酸异山梨酯缓释片对慢性高原病大鼠模型睾丸的保护作用

Protective effects of Isosorbide-5-Mononitrate Sustained Release Tablets on the testis of chronic mountain sickness rat model

目的研究单硝酸异山梨酯缓释片对慢性高原病(CMS)大鼠模型睾丸的保护作用。方法建立CMS模型,随机分为正常对照组、高原模型组、阳性对照组和单硝酸异山梨酯缓释片低、中、高剂量组。观察其肺动脉压(PAP)和睾丸组织病理学的变化,测定白介素-6(IL-6)、C-反应蛋白(CRP)、超氧化物歧化酶(SOD)、丙二醛(MDA)和谷胱甘肽-过氧化氢酶(GSH-PX)等指标。结果与正常对照组比较,高原模型组、阳性对照组和单硝酸异山梨酯缓释片各剂量组大鼠体内的PAP、IL-6、CRP和MDA水平升高,SOD和GSH-PX水平降低,光镜下可见大鼠睾丸组织受到损伤,提示低氧对大鼠的睾丸造成损害。与高原模型组比较,单硝酸异山梨酯缓释片高剂量组大鼠体内的PAP、IL-6、CRP和MDA水平降低,SOD和GSH-PX水平升高,均接近正常值,且优于阳性对照组。同时,睾丸组织病理切片亦出现相应改善。结论高剂量的单硝酸异山梨酯缓释片对CMS大鼠模型睾丸有保护作用。

To study the role of Isosorbide-5-Mononitrate Sustained Release Tablets on the protection effect in the testis of the chronic mountain sickness rat model.Rats were randomly divided into blank control group,high altitude group,nifedipine group,and low,medium,high dosage Isosorbide-5-Mononitrate Sustained Release Tablets group.The changes of pulmonary artery pressure(PAP)and the testis tissue were observed.The test was conducted in these rats to check their interleukin-6(IL-6),C-reactive protein(CRP),superoxide dismutase(SOD),malondialdehyde(MDA)and glutathione peroxidase(G SH-PX).Compared with the blank control group,the PAP,IL-6,CRP and MDA levels of high altitude group,nifedipine group,high,medium,low dosage Isosorbide-5-Mononitrate Sustained Release Tablets group were higher,but SOD and GSH-PX levels were lower.Optical microscopy showed that testis tissue was injured,which indicated the testis of the rat was damaged under hypoxic conditions.Compared with the high altitude group,the PAP,IL-6,CRP and MDA levels of high altitude group,nifedipine group,and high,medium,low dosage Isosorbide-5-Mononitrate Sustained Release Tablets group were lower,but the SOD and GSH-PX levels were higher.Each index was closed to the normal result and better than the positive control group index.The testis tissue pathology also appeared satisfactory results.High dosage Isosorbide-5-Mononitrate Sustained Release Tablets has protection effect in the testis of the chronic mountain sickness rat model.
