摘要
随着家蚕抗血液型脓病新品种‘华康’系列在全国的推广和应用,市面上出现了一些鱼龙混杂现象。针对这一问题,本研究用SLAF-seq(Specific-locus amplified fragment sequencing)技术对15个不同的家蚕品种(7个抗脓病品种和8个非抗脓病品种)进行简化基因组测序,结果共获得所有检测品种的447 359个SNP位点,分析这些SNP标记在家蚕的28条染色体上的分布以及与抗脓病性能的相关性,得到第27群上的50个与抗BmNPV相关的SNP分子标记;经过PCR验证的结果与SLAF-seq测序结果一致,认为这些SNP可以作为鉴定和判别‘华康’系列品种的分子标记。
With the promotion and application of new silkworm varieties‘HuaKang’series in China,all kinds of silkworm production flocked to join,and the bad became mixed with the good,finally damage to the interests of farmers and breeders.To solve this problem,Specific-Locus Amplified Fragment Sequencing(SLAF-Seq)technology was carried out on 15 different silkworm varieties(7 of 15 are resistant to BmNPV,while the rest 8 varieties are susceptible to BmNPV).447 359 SNPs,which distributed on 28 chromosomes,were detected from all samples after mapped to the reference genome.Bioinformatics analysis of all SNPs showed that 50 SNPs located on chromosome 27 may correlated to the BmNPV resistance.PCR validation were then performed and showed the consistent with the SLAF-Seq results.The present identified SNPs can be the molecular markers used to identify and discriminate of the‘Huakang’series from the market
