Box-Behnken效应面法优化半枝莲和白花蛇舌草药对中总黄酮的提取工艺

Optimization of conditions for extraction of total glucoside in Hedyotic diffusa and Scutellaria barbata using Box-Behnken response surface

目的优选半枝莲和白花蛇舌草药对中总黄酮的提取工艺。方法以配比、乙醇体积分数、料液比、提取时间为考察因素,采用Box-Behnken效应面法,以总黄酮提取率为考察指标,优化其提取的工艺条件,并以芦丁为对照品,三氯化铝为显色剂,采用紫外分光光度法测定药对中黄酮的含量。结果确定最佳的提取工艺条件为半枝莲和白花蛇舌草配比为2.85∶1,乙醇体积分数为65.15%,料液比为1∶28.09,回流提取2次,提取时间为2.49h。结论 Box-Behnken效应面法优选的提取工艺简便可行,预测效果好,可以得到较高含量的黄酮,有一定的应用前景。

Objective:To optimize the extracting process of total flavonoids in Drug pairs.Method:Box-Behnken response surface was employed with the ratio of drug pairs,the ethanol concentration,the ratio of solid-liquid,the extraction time as factors.The response index was the extraction rate of total flavonoids.UV-Vis spectroscopy was applied to determine the content of total flavonoids in drug pairs.Result:Optimum extracting conditions was as follows:65.15%ethanol,28.09 times and2.49 hour respectively.Conclusion:The optimized extraction method is simple,feasible and has a good application prospect.The higher total flavonoids content can be obtained in this condition.
