摘要
通过能量转移建立了溶菌酶荧光猝灭法测定槲皮素。荧光猝灭法研究了不同外界因素如溶菌酶浓度、最佳反应时间、pH及温度对槲皮素猝灭溶菌酶荧光的影响。淡竹叶、夏枯草提取液对溶菌酶的荧光均可产生强猝灭作用,槲皮素对溶菌酶荧光猝灭呈线性关系;线性范围为8×10^(-7)~2×10^(-5)mol·L^(-1),相关系数R^2=0.9936检出限为1.31×10^(-6)mol·L^(-1)。并对夏枯草、淡竹叶两种样品黄酮含量进行分析。
A fluorescence quenching method was established for the determination of quercetin by energy transfer.Fluorescence quenching was used to study the effects of different external factors such as lysozyme concentration,optimal reaction time,pH and temperature on the quenching fluorescence of lysozyme.The results showed that the extract of Lactuca sativa and Prunella vulgaris had a strong quenching effect on the fluorescence of lysozyme and the linear relationship of quercetin to the fluorescence quenching of lysozyme.The linear range was 8×10-7~2×10-5 mol·L-1,the correlation coefficient R 2=0.9936,the detection limit was 1.31×10-6 mol·L-1.The contents of flavonoids in Prunella vulgaris L and lophantherum gracile brongn were analyzed.
