摘要
目的:观察S100钙结合蛋白A4(S100A4)和骨桥蛋白(OPN)在糖尿病大鼠视网膜病变中的表达,探讨S100A4和OPN在糖尿病视网膜病变诊断和治疗中的意义。方法:选取健康雄性SD大鼠36只,用抛硬币法随机选取18只进入模型组,另18只进入对照组。模型组一次性腹腔注射链脲佐菌素(STZ)45mg/kg,对照组一次性腹腔注射同体积的柠檬酸缓冲液。注射3d后于尾静脉采血测血糖浓度,连续3次>16.65mol/dl,且尿糖阳性为造模成功。分别于造模成功后4周、12周、20周随机处死各组大鼠6只,立即取其双眼眼球。ELISA法检测视网膜S100A4的浓度,real-time PCR法检测OPN mRNA的水平。所得数据组间比较分别采用析因分析,组内两两比较分别采用LSD检验,P<0.05为差异有统计学意义。结果:与同期对照组比较,4周、12周、20周模型组的大鼠视网膜S100A4和OPN mRNA表达量均增高,随着时间延长有增高的趋势,差异有统计学意义(F=1845.893,P<0.001;F=3650.307,P<0.001)。结论:S100A4和OPN是糖尿病视网膜病变新的诊断标志物和治疗靶点。
Objective:To observe the expression of S100A4 and OPN in rats with diabetic retinopathy,and explore the significance of S100A4 and OPN in the diagnosis and treatment of diabetic retinopathy.Methods:36 healthy male SD rats were selected,and 18 rats were randomly selected into the model group by coin tossing method.The other 18 rats were included in the control group.The model group was established by one-time intraperitoneal injection of STZ 45mg/kg,and the control group was injected intraperitoneally with citric acid buffer of the same volume.After 4 weeks,12 weeks and 20 weeks,6 rats in each group were killed at random,and the eyeball was taken immediately.The concentration of S100A4 in retina was detected by ELISA,and the level of OPN mRNA was detected by real-time PCR.Results:Compared with the control group at the same time,the expressions of S100A4 and OPN mRNA in the retina of diabetic rats in the model group were increased at 4,12 and 20 weeks.There was a tendency to increase as time went on,with a statistically significant difference(F=1845.893,P<0.001;F=3650.307,P<0.001).Conclusion:S100A4 and OPN are new diagnostic markers and therapeutic targets of diabetic retinopathy.
