摘要
目的:观察BN大鼠脉络膜新生血管(CNV)模型视网膜和脉络膜中二酰基甘油(DAG)、活化蛋白激酶C(PKC)表达的变化。方法:将12只BN大鼠分为正常组、模型组,各6只,模型组采用532nm激光光凝建立BN大鼠CNV模型,造模成功后每周行荧光素眼底血管造影(FFA)、光学相干断层扫描(OCT)等检查,于造模后5周处死动物,并行酶联免疫吸附测定法检测视网膜及脉络膜中DAG、PKC的表达水平。结果:造模5周后FFA及OCT检测可见CNV形成,酶联免疫吸附测定法检测模型组视网膜及脉络膜上DAG、PKC表达水平均上调,与正常组比较,差异有统计学意义(P<0.05)。结论:532nm激光光凝建立的BN大鼠CNV模型可作为进一步研究CNV的动物模型,通过酶联免疫吸附测定法检测其视网膜及脉络膜内DAG、PKC的含量与BN大鼠模型CNV的形成有相关性。
Objective:To observe the changes of the expression of retinal and choroid diacylglycerol and protein kinase C in choroidal neovascularization model of brown norway rats.Methods:12 BN rats were randomly divided into 2 groups,namely the normal group and model group.CNV model of brown norway rats in the model group was established by 532nm laser photocoagulation.Fundus fluorescein angiography and optical coherence tomography were performed per week.In 5 weeks,the rats were sacrificed and the expression of retinal and choroid DAG,PKC was detected by ELISA assay.Results:In 5 weeks,CNV was formed via FFA and OCT in the model group and retina and choroid DAG and PKC were highly raised;the difference between groups was statistically significant(P<0.05).Conclusions:CNV model of brown norway rats which was established by 532nm laser photocoagulation could be selected as an animal model for further study on CNV,and the content of DAG and PKC in the retina and choroid is associated with the form of CNV.
