摘要
目的探讨外源性硫化氢对大鼠精索静脉曲张(VC)生精功能损伤的保护作用。方法选取成年雄性SD大鼠36只,随机分为假手术组、模型组和外源性硫化氢组。外源性硫化氢组在VC模型成功建立基础上注射外源性H_2S[20 mg/(kg·d)]。采用HE染色观察睾丸组织病理改变;测定睾丸组织中丙二醛(MDA)、超氧化物岐化(SOD)含量;采用原位缺口末端标记检测细胞凋亡指数;免疫组化检测睾丸组织中Bax和Caspase-3的表达情况;RT-PCR检测Bax和Caspase-3 mRNA表达水平。结果模型组可见各级生精细胞排列紊乱且广泛脱落等表现,Bax及Caspase-3表达、氧化应激水平和凋亡指数明显高于假手术组;与模型组比较,外源性硫化氢组Bax、Caspase-3表达水平及氧化应激水平和凋亡指数明显降低,各组间比较差异均有统计学意义(P<0.05)。结论外源性硫化氢可减轻氧化应激水平和抑制凋亡,从而对VC睾丸生精功能发挥保护作用。
Objective To investigate the protective effects of hydrogen sulfide on spermatogenic function injury in rats with varicocele(VC).Methods Thirty-six adult male SD rats were randomly divided into 3 groups,with 12 rats in each group:sham group,VC group and exogenous hydrogen sulfide group.Hydrogen sulfide[20 mg/(kg·d)]was injected intraperitoneally on the basis of the successful establishment of VC model.HE staining was used to observe the pathological changes of testis;contents of malondialdehyde(MDA)and superoxide dismutase(SOD)were determined;cell apoptosis index was detected by dUTP nick end labeling;the expression of Bax and Caspase-3 in testis tissue was detected by immunohistochemistry;mRNA expression of Bax and Caspase-3 in testis tissue was measured by RT-PCR.Results The left testicular tissue showed desquamation of epithelial cells and disordered arrangement of spermatogenic cells in VC group.The levels of Bax and caspase-3,oxidative stress and apoptosis index were significantly higher than those in the sham group;compared with VC group,the levels of Bax and caspase-3,the oxidative stress and apoptosis index significantly decreased in the hydrogen sulfide group(P<0.05).Conclusion Hydrogen sulfide reduces the level of oxidative stress and inhibits the cell apoptosis.Thus,it plays a protective role in spermatogenic function injury in varicocele rats.
