摘要
建立白花龙胆的胶束毛细管电泳指纹图谱,为白花龙胆质量控制提供依据.以50mmol/L硼砂-60mmol/L十二烷基磺酸钠(SDS)-20%的乙腈(pH=9.44)为运行缓冲液,未涂渍的熔融石英毛细管(64.5cm×50μm,有效长度56cm)为分离通道,在分离电压18kV,压力进样5kPa×5s,检测波长254nm的电泳条件下测定青藏高原9个地区的10种白花龙胆.结果表明:选择异荭草苷峰作为参照物峰,建立有12个共同峰的胶束毛细管电泳指纹图谱.该方法稳定性和重现性好,测得白花龙胆药材指纹图谱具有一定的特征性和专属性.
To establish high performance capillary electrophoresis fingerprint of Gentiana algida Pall.,a basis has been provided for the quality control of Gentiana algida Pall..50mmol/L borax-60mmol/L sodium dodecyl sulfonate-20%acetonitrileas running buffer(pH=9.44)no coating standard fused silica capillary(64.5 cm×50μm,effective length of 56 cm)for separation channel.At the separation voltage is 18 kV,injection pressure is 50 mbar x 5 s,and detection wavelength is 254 nm,determination 10 kinds of Gentiana algida Pall.in 9 different areas of Qinghai Tibet Plateau.It is concluded that Chromatographic peaks of Isoorientin as reference peak,building with 12 common peaks of micelle capillary electrophoresis fingerprint.The method with good stability and good reproducibility,high performance capillary electrophoresis fingerprint of Gentiana algida Pall.has certain characteristic and specificity.
