摘要
目的探讨低氧对肺癌细胞系A549增殖和侵袭的影响及其可能的分子机制。方法将体外培养的非小细胞肺癌细胞系A549分别置于常氧和低氧环境下刺激24 h,采用CCK8法检测细胞的增殖能力;Transwell侵袭实验检测细胞侵袭能力;Real time-PCR和Western blot法检测细胞中Wnt/β-catenin mRNA和蛋白的表达水平;并进一步使用β-catenin特异性siRNA阻断其表达,并检测。结果体外低氧微环境可显著促进肺癌细胞系A549的增殖和侵袭,并可激活Wnt/β-catenin信号通路;而抑制Wnt/β-catenin信号通路则能有效抑制低氧介导的肺癌细胞增殖与侵袭。结论低氧微环境通过激活Wnt/β-catenin信号通路促进非小细胞肺癌细胞系A549的增殖和侵袭转移。
Objective To study the effect of hypoxia on proliferation and invasion in non-small-cell lung cancer cell line A549 and its molecule mechanism.Methods The non-small-cell lung cancer cell line A549 were stimulated under hypoxia and normoxia for 24 hours in vitro.The proliferation ability was detected by CCK8.The invasion ability was detected by Transwell assay.The mRNA and protein expression levels of HIF-1αandβ-catenin were detected by RT-PCR and Western blot.Furthermore,β-catenin siRNA was used to investigate the role ofβ-catenin on hypoxia induced proliferation and invasion.Results The results demonstrated that hypoxia could promote A549 cells proliferation and invasion.The mRNA and protein level of HIF-1αandβ-catenin was upregulated by hypoxia treatment,whereasβ-catenin siRNA could block these processes.Conclusion Hypoxia can promote the proliferation and invasion of non-small-cell lung cancer cell line A549 by activating Wnt/β-catenin signaling pathway.
