摘要
为监测新疆地区自然环境中蓝舌病流行情况,并分离蓝舌病病毒(BTV),本研究在新疆巴州尉犁县设置10只山羊、10只绵羊为哨兵动物,自2016年5月-11月期间每周采集哨兵动物的血清和抗凝血,对所有样品进行BTV抗体检测,并将阳性及转阳前、后一周的抗凝血样本接种于BHK-21细胞,盲传5代后有14份细胞出现病变,对这些病变细胞液进行Ac-ELISA检测、BTV VP7基因PCR鉴定。结果显示BTV抗原检测均为阳性,经RT-PCR扩增,在1 156 bp处均获得目的基因片段。本研究在新疆成功分离到了羊源BTV。
To monitor the prevalence of bluetongue disease in natural environment of Xinjiang and to isolate BTV,10 goats and 10 sheep were selected as sentinel animals in Yuli County,Bazhou.From May to November,2016,serum and anticoagulant of sentinel animals were collected weekly,all samples were tested for BTV antibody,and anticoagulant samples of positive and the ones with one week before and after turning to positive were vaccinated into BHK-21 cells,after blind passages of 5 generations,lesions on 14 cells were observed.The pathological cell fluid was tested by Ac-ELISA and identified by BTV VP7 gene PCR.The results showed that all the BTV antigens were positive,and target gene fragment was obtained at 1 156 bp by RT-PCR.In this study,sheep-derived BTV was successfully isolated in Xinjiang.
