聚山梨酯80修饰的天麻素脂质体制备研究 被引量:1

摘要 目的:聚山梨酯80(polysorbate-80,P-80)修饰的天麻素脂质体制备工艺研究。方法:采用星点设计-效应面法优化处方,以脂质浓度、磷脂与胆固醇比例、脂质与药物比例为考察因素,包封率和载药量为评价指标,预测最佳处方进行验证并对脂质体进行表征。结果:最优处方为:脂质浓度16mg/ml,卵磷脂:胆固醇=3:1,脂质:天麻素=5:1;依照最优处方制备的脂质体呈类球形,平均粒径为(97.63±1.53)nm、多分散系数为(0.27±0.02)、电位为(-14.1±0.83)mv、包封率与载药量分别为(35.17±0.25)%、(7.03±0.05)%。结论:薄膜-超声法制备脂质体的工艺简单易行;P-80修饰的天麻素脂质体呈淡蓝色,结果表明其外观呈类球形、性质稳定,为其进一步的体内研究奠定基础。 Objective:To optimize the formulation of the gastrodin liposomes modified with polysorbate 80 with thin film ultrasonic method by Box-Behnken design and response surface method.Methods:The influence factors included the concentration of lipid,the ratio of phosphatidylcholine to cholesterol,the ratio of lipid to drug.The evaluation parameters included entrapment efficiency,drug loading.Experiments were done on the theory of Box-Behnken design.The data were simulated and the possibly optimal formulation was predicted by response surface method.Results:The optimal formulation were as follow as the concentration of lipid was 16 mg/ml,phosphatidyl choline-cholesterol ratio of 3.05:1,lipid-drug ratio of 5:1.According to the optimal conditions,the mean diameter,polydis persity index,zeta potential of the liposome were(97.63±1.53)nm,(0.27±0.02)and(-14.1±0.83)mv,entrapment efficiency,drug loading were(35.17±0.25)%and(7.03±0.05)%,respectively.Conclusion:The preparation of liposomes bythin film ultrasonic method is simple and feasible.The gastrodin liposomes modified with polysorbate 80 were pale blue.The characterization results showed that the liposomes were spherical in shape and stable in nature,which lay the foundation for further research on in vivo research.
