摘要
针对10份不同产地的铁皮石斛,采用甲醇超声萃取,Oasis HLB固相萃取柱净化,Hypersil BDS C_(18)色谱柱分离,二极管列阵检测器测定,建立高效液相色谱法(high performance liquid chromatography,HPLC)同时测定13种酚酸类物质的检测方法。结果表明:在0.5μg/mL^50.0μg/mL范围内,具有良好的线性关系(R^2≥0.999 6);LOD在0.07 mg/kg^0.3 mg/kg之间;LOQ在0.2 mg/kg^1.0 mg/kg之间;加标回收率在86.7%~102.3%之间,RSD在1.1%~3.5%之间;铁皮石斛中共检出12种酚酸,阿魏酸含量最高,其次是丁香酸、二氢阿魏酰酪胺、对羟基苯甲酸,这些酚酸可能与铁皮石斛的抗氧化、增强免疫力、降血糖等功效有关,这将为铁皮石斛活性物质研究与开发提供参考。
For 10 Dendrobium officinale from different origins,the determination of 13 kinds of phenolic acids by high performance liquid chromatography(HPLC)was carried out using ultrasonic extraction with methanol,purification with Oasis HLB solid phase extraction column,Hypersil BDS C18 column separation and diode array detector.Material testing methods.The results showed that there was a good linear relationship(R2逸0.999 6)in the range from 0.5滋g/mL to 50.0滋g/mL,the LOD were range from 0.07 mg/kg to 0.3 mg/kg and the LOQ were range from 0.2 mg/kg to1.0 mg/kg.The recoveries were range from 86.7%to 102.3%,RSD were range from 1.1%to 3.5%.12 kinds of phenolic acids were detected in Dendrobium officinale,and the content of ferulic acid was the highest,followed by syringic acid,dihydro ferulic acid tyramine,p-hydroxybenzoic acid,and these phenolic acids may be antioxidants,enhance immunity,hypoglycemic effect,which will provide refer原ence for the research and development of active substance of Dendrobium officinale.
