摘要
优化北虫草酶活测定条件,为北虫草高产菌株的筛选和品种鉴定提供参考,利用单因素和响应面法,从pH、温度及底物浓度3个方面研究北虫草中纤维素酶活性测定的最适方法。结果表明:北虫草中纤维素酶活性测定的最佳条件是温度50℃、pH 7.0和酶液量0.9mL,酶活最高值为0.138 04U/g。
In order to optimize the conditions for determination of Cordyceps militaris cellulase and provide references for high-yield strains screening and cultivars identification,the authors studied the most suitable conditions for the determination of cellulase activity in C.militaris from three aspects of pH,temperature and substrate concentration by single-factor and response surface method.The results showed that the optimum conditions for the determination of cellulase activity were 50℃,pH 7.0,enzyme volume 0.9 mL.The highest enzyme activity value is 0.138 04 U/g.
