摘要
采用间苯三酚-浓硫酸显色法,建立双波长分光光度法测定溶液中木糖和葡萄糖含量,并对最佳测定条件及溶液中木糖及葡萄糖共存时的相互影响进行了分析,探究其测定木质纤维素酶解液中木糖和葡萄糖含量的可行性。实验结果表明:与间苯三酚-浓硫酸显色后,木糖和葡萄糖的最大吸收峰分别为444 nm和420 nm;最佳测定条件为:1.0 m L木糖或葡萄糖标准溶液,加5.0 m L显色剂,100℃水浴加热10 min。木糖和葡萄糖浓度在20~120μg/m L范围内线性关系良好;精密度实验得到木糖和葡萄糖的标准偏差分别为0.8432和0.8430μg/m L,RSD分别为1.3967%和1.3959%;加标回收实验得到木糖和葡萄糖的回收率分别为100.82%和100.65%,RSD分别为0.32%和0.46%。该方法操作简单,精确度高,可用于实验室快速测定溶液中葡萄糖和木糖的含量,但两种单糖共存时对测定结果有一定的影响。
The concentration of xylose and glucose in solution were determined simultaneously by phloroglucinol colorimetry.The optimal determination conditions,the interaction between two monosaccharides and its application were studied.The results showed that the maximum absorption peak of xylose and glucose was observed at the wavelength of 444 nm and 420 nm,respectively.Optimal determination conditions were as follows:1 mL substrates with 5 mL coloring agent,boiling water bath for 10 min.Results showed that both monosaccharides obeyed Beer-Lambert’s law at the concentrations of 20~120μg/mL.The experiment indicated that the standard deviation of xylose and glucose were 0.8432 and 0.8430μg/mL,RSD were 1.3967%and 1.3959%,respectively.Recovery rate was 100.82%and 100.65%,RSD were 0.32%and 0.46%,respectively.This method was simple and accurate and had high application in the rapid determination of glucose and xylose.However,the interaction between the two monosaccharides limited its application when two monosaccharides coexisted.
