摘要
目的:检测谷氨酰胺酶1(GLS1)在结肠癌和癌旁组织中的表达,并观察干涉GLS1对结肠癌细胞代谢重编程的影响。方法:利用免疫组织化学实验观察50例结肠癌组织芯片中GLS1的表达状态,并进行统计学量化分析;利用siRNA干涉HT29细胞和Caco2细胞中的GLS1后,通过MTT实验、葡萄糖摄取实验和谷氨酰胺摄取实验检测其对细胞生物学行为的影响。结果:GLS1在结肠癌中显著高表达;干涉GSL1后,细胞增殖能力、对葡萄糖和谷氨酰胺的摄取能力均显著下降。结论:GLS1在结肠癌组织中高表达,并且影响细胞对葡萄糖和谷氨酰胺的摄取能力。
Objective:To determine glutaminase 1(GLS1)expression in colorectal cancer and adjacent normal tissues,and detect GLS1 knockdown effect on colorectal cancer metabolic reprogramming.Methods:GLS1 expression was determined in 50 colorectal cancer tissues and adjacent normal tissues.The statistical analysis was further performed.In HT29 and Caco2 cells after GLS1 knockdown with specific siRNA,GLS1 effects on cell behaviors were further detected through MTT assay,glucose uptake assay and glutamine utilization assay.Results:GLS1 is highly expressed in colorectal cancers.The reduction of GLS1 by siRNA decreased cell growth rate and the capacities of glucose and glutamine utilizations.Conclusion:GLS1 is highly expressed in colorectal cancers and plays a key role in cell glucose and glutamine utilizations.
