CXCL1促进人肝癌细胞发生内质网应激

摘要 目的:探究趋化因子CXCL1过表达对人肝癌HepG2细胞内质网应激的影响。方法:将GFP-CXCL1质粒转染人肝癌HepG2细胞,荧光显微镜观察绿色荧光蛋白;将该质粒转染人肝癌HepG2细胞,24 h后收集细胞,提取细胞总mRNA后进行反转录,荧光定量PCR检测内质网应激相关蛋白IRE1、PERK、ATF6的表达。结果:CXCL1过表达后,通过荧光定量PCR检测出内质网应激相关蛋白IRE1、PERK、ATF6均升高。结论:过表达的CXCL1能够促进人肝癌HepG2细胞的内质网应激,为后续研究奠定了基础。 Objective:To investigate the effect of CXCL1 overexpression on endoplasmic reticulum stress in human hepatoma HepG2 cells.Methods:GFP-CXCL1 plasmid was transfected into HepG2 cells,and the expression of green fluorescent protein was observed by fluorescence microscope.The plasmid was transfected into HepG2 cells.After 24 hours,cells were harvested and total cellular mRNA was extracted.Reverse transcription was performed.Finally,the expression of endoplasmic reticulum stress-related proteins IRE1,PERK and ATF6 were detected by real-time quantitative PCR.Results:After CXCL1 was overexpressed,the endoplasmic reticulum stressrelated proteins IRE1,PERK and ATF6 were detected by fluorescence quantitative PCR.Conclusion:Overexpression of CXCL1 can promote the endoplasmic reticulum stress in human hepatoma HepG2 cells,which lays the foundation for further research.
