富硒荷叶离褶伞菌丝体中硒多糖提取工艺的优化及红外光谱分析 被引量:11

摘要 以20 L生物罐发酵培养的富硒荷叶离褶伞菌丝体为原料,优化菌丝体中硒多糖的提取工艺并通过红外光谱对水溶性硒多糖的结构进行分析。通过单因素试验和Box-Behnken试验设计方法得到硒多糖的最佳提取工艺条件:提取时间为50 min、提取温度83℃、料液比1∶130(g∶m L)、提取次数2次。测得多糖提取率为44.62%,较预测值增加0.02%,此时多糖中硒含量达到31.9μg/g。将纯化后的硒多糖进行红外光谱分析,富硒未改变水溶性硒多糖的主体结构,但吡喃环的吸收峰发生了改变,说明在荷叶离褶伞菌丝体富硒培养过程中,硒参与了硒多糖的合成。 Selenium-enriched of Lyophyllum decastes mycelium was cultured in 20L fermenter as raw material in this study.The extraction process of selenium polysaccharide from mycelium of Lyophyllum decastes was optimized and the structure of refined polysaccharides were characterized by IR spectroscopy.One-factors-at-a-time experiments and Box-Behnken experimental design coupled with response surface methodology were carried out to optimize the process parameters of Se-polysaccharides.The results showed that the optimum conditions were:50 min at 83℃,the ratio of solid to liquid 1∶130(g∶mL),and two times extraction.Under the above conditions,the extraction rate of polysaccharide was 44.62%,which was 0.02%more than predicted,and the selenium content achieved 31.9μg/g in polysaccharide.After purifying,the refined polysaccharides were characterized by IR spectroscopy.The result showed that the water soluble polysaccharides were pyranopolysaccharide linked byβ-glycosidic.The primary structure of water-soluble polyaccharides was not changed,but absorption peaks of pyranring was changed.This further experiment demonstrated that Se participated in the synthesis of Se-polysaccharides during the cultivation of Se-enriched Lyophyllum decastes mycelium.
