共表达猪IL-2与融合抗菌肽重组质粒的构建及其小鼠免疫效应研究 被引量:1

摘要 为研制新型高效安全的动物免疫调节剂,本实验采用In-fusion克隆技术、2Aself-cleavage技术构建pVAX1融合抗菌肽与白细胞介素2基因真核共表达载体:CAM PS-2A-IL2(VAP2)。构建的重组质粒用壳聚糖包裹并转染HEK293细胞,通过猪外周血淋巴细胞刺激实验检测表达上清活性。将包裹的重组质粒纳米颗粒腹腔注射接种4周龄的雌性昆明小鼠作体内生物学活性研究。并于28d后用大肠杆菌(Amp、Kana高耐受)和金黄色葡萄球菌(Amp、Kana高耐受)进行攻毒试验,观察每组小鼠的发病率和死亡率。实验结果表明VAP、VAP2两实验组CD^(4+)、CD^(8+)细胞,IgG,IgG1,IgG2a含量以及TLR s,IFN-r等免疫相关因子的表达水平均明显高于对照组pVAX1(P<0.05)。而且两实验组相比较,VAP2组明显高于VAP组(P<0.05)。以上实验结果表明,CS包裹的VAP,VAP2能有效防止病原微生物感染小鼠,增强机体的体液免疫和细胞免疫能力,并且VAP2的作用要明显优于单一抗菌肽融合基因,表明抗菌肽融合基因与白细胞介素2的共表达对动物有更好的保护效应,这为研究新型有效的免疫分子以增强机体对感染的抵抗力奠定了基础,具有重要的学术价值和应用。
