胃癌组织中COX-2、NF-κB及VEGF的表达及与血管生成的关系 被引量:13

Expression of COX-2,NF-κB and VEGF in gastric carcinoma tissues and its relationship with angiogenesis

摘要 目的观察胃癌组织中环氧合酶-2(COX-2)、细胞核因子-κB(NF-κB)、血管内皮生长因子(VEGF)的表达情况及与胃癌患者临床病理特征和胃癌组织中肿瘤血管新生的关系。方法采用免疫组织化学法检测69例胃癌患者癌组织及其中40患者癌旁组织中COX-2、NF-κB、VEGF表达情况,分析各指标与患者临床病理特征的关系;另以抗CD34抗体标记组织标本中的微血管内皮细胞,计算并比较COX-2、NF-κB、VEGF不同表达情况的胃癌患者微血管密度(MVD)。结果 COX-2及VEGF主要表达于细胞质及细胞膜中,而NF-κB主要表达于细胞核及细胞质中,且胃癌组织中COX-2、NF-κB、VEGF阳性着色更为明显;不同性别、年龄、肿瘤大小的胃癌患者胃癌组织中COX-2、NF-κB、VEGF阳性表达率比较差异均无统计学意义(P均>0.05);淋巴结转移者COX-2、NF-κB阳性表达率显著高于淋巴结未转移者(P均<0.05),而VEGF阳性表达率差异无统计学意义(P>0.05);胃癌组织中COX-2、NF-κB、VEGF阳性表达者MVD均显著高于阴性表达者(P均<0.05),且三者均阳性表达者MVD显著高于三者中任意一个阴性表达者(P<0.05)。结论胃癌组织中COX-2、NF-κB、VEGF阳性表达率显著高于癌旁组织,三者均可明显促进胃癌组织中肿瘤血管新生。 Objective:It is to observe the expression of circulating oxygenase-2(COX-2),nuclear factor-κB(NF-κB)and vascular endothelial growth factor(VEGF)in gastric cancer and its relationship with the clinicopathological characteristics and angiogenesis in gastric cancer.Methods 0x09 The expression of COX-2,NF-κB and VEGF in cancer tissues of 69 patients with gastric cancer and 40 cases of paracancerous tissue from them were detected by immunohistochemistry.The relationship between the indexes and the clinicopathological features of the patients was analyzed.microvascular density(MVD)in gastric cancer patients with different expressions of COX-2,NF-κB and VEGF were calculated and compared by labeled the microvascular endothelial cells in tissue specimens with anti CD34 antibody.Results 0x09 COX-2 and VEGF were mainly expressed in cytoplasm and cell membrane,and NF-κB was mainly expressed in the nucleus and cytoplasm,the positive staining of COX-2,NF-κB and VEGF in gastric carcinoma were more obvious.There were no significant different in the positive expression rates of COX-2,NF-κB and VEGF of gastric cancer tissues among different gender,age and tumor size(all P>0.05).The positive expression rates of COX-2 and NF-κB in patients with lymph node metastases were significantly higher than those without metastases(all P<0.05),there was no significant difference in the positive expression rate of VEGF(P>0.05).The MVD in the gastric cancer tissues with positive expressions of COX-2,NF-κB and VEGF were significantly higher than with negative expressions(all P<0.05),the MVD with all positive expression was significantly higher than that of any negative expression in them(P<0.05).Conclusion 0x09 The positive expression rate of COX-2,NF-κB and VEGF in gastric carcinoma is significantly higher than that in para cancerous tissue,and all them can obviously promote the neovascularization of tumor in gastric cancer tissue.
