中药调节肿瘤细胞内质网应激性凋亡

摘要 内质网是细胞中重要的细胞器之一,主要负责蛋白质的合成、转运、信号转导和钙离子调节等功能,参与新生成蛋白多肽的折叠修饰并保证新生成蛋白多肽折叠的正确、顺利进行。多种生理或病理条件下,如当细胞处于持续的化学刺激、氧化应激、钙离子或者能量代谢失衡时,细胞内环境稳态将被破坏,内质网功能发生紊乱,主要表现为二硫键不能形成,引起蛋白质的错误折叠及蛋白质在内质网腔内积聚,即内质网应激(endoplasmic reticulum stress,ERS)[1]。内质网应激可以分为两个阶段,早期的非折叠蛋白反应及晚期的诱导凋亡。即适当的内质网应激通过触发非折叠蛋白反应,从而使受损的细胞存活,相反,持续的内质网应激则会引起细胞凋亡[2]。
