响应面优化杜仲提取物抗氧化活性及其工艺研究 被引量:7

摘要 目的:研究响应面优化后的杜仲提取物对线虫寿命的影响。方法:以对羟基自由基、超氧去离子自由基、DPPH自由基的清除能力为指标,通过响应面分析法对杜仲提取物进行工艺优化,优化后的杜仲提取物进行线虫寿命实验,验证其抗氧化、抗衰老效果。结果:优化后的提取条件为提取温度70℃、提取时间2 h、溶剂体积8 mL·g-1。线虫寿命实验证明,杜仲给药组的线虫平均寿命为24.95 d,远大于对照组和总黄酮给药组。结论:该工艺操作简单、耗能少,线虫实验为杜仲提取物的抗氧化活性提供了更有力的依据,为杜仲的进一步开发和利用提供了参考。 Objective:To study the effect of Eucimmia ulmoides extract optimized by response surface on life of nematode.Methods:Taking the scavenging ability of hydroxyl radical,superoxide radical,free radical and DPPH radical as indexes,the extraction process of E.ulmoides extract was optimized by response surface methodology.The optimized E.ulmoides extract was used to test its antioxidant and anti-aging effects.Results:The optimized extraction conditions were as follows:extraction temperature 70℃,extraction time 2 h,and solvent volume 8 mL·g-1.Nematode life test showed that the average life of nematodes in E.ulmoides injection group was 24.95 days,which was much higher than that of control group and total flavone group.Conclusion:The process is simple and less energy consuming,and the nematode test provides a more powerful basis for the antioxidant activity of E.ulmoides extract,which provides a reference for further development and utilization of E.ulmoides.
