LC-MS/MS法同时测定十一味益肾降糖片中马钱苷和莫诺苷的含量

Simultaneous Determination of Loganin and Morroniside in Eleven Ingredient Yishen Jiangtang Tablet by LC-MS/MS

目的:采用高效液相色谱-串联质谱法(LC-MS/MS)技术建立同时测定十一味益肾降糖片中马钱苷和莫诺苷含量的方法。方法:以水-乙腈溶液为流动相梯度洗脱,以Thermo Scientific Hypersil ODS色谱柱为分析柱,通过电喷雾离子源,正离子模式,选择离子监测。结果:马钱苷和莫诺苷的线性范围均为10~2000 ng·mL^(-1),相关系数分别为0.998 2和0.999 1,加样回收率在97.28%~100.41%,RSD均低于1.10%。结论:通过LC-MS/MS技术建立的十一味益肾降糖片中马钱苷和莫诺苷的含量测定方法专属性强,快速、灵敏、准确,可用于其质量控制。

Objective:To develop a LC-MS/MS method for the determination of loganin and morroniside in Eleven Ingredient Yishen Jiangtang Tablet.Methods:The gradient elution was carried out with mobile phase consisting of water-acetonitrile.The separation was performed on a Thermo Scientific Hypersil ODS column and the mass spectrometer was operated in the positive ionization electrospray mode using selective reaction monitor for analysis of two components.Results:The linear ranges of loganin and morroniside were both 10-2000 ng·mL-1,and the linear correlation coefficients were 0.998 2 and 0.999 1,respectively.The recoveries of the analytes ranged from 97.28%-100.41%and the relative standard deviations were all less than 1.10%.Conclusion:The determination of loganin and morroniside in Eleven Ingredient Yishen Jiangtang Tablet by LC-MS/MS is high specific,rapid,sensitive and accuracy and the method could be applied for the quality control of Eleven Ingredient Yishen Jiangtang Tablet.
