摘要
[目的]建立变异冬红果初代培养无菌体系。[方法]以变异冬红果茎段为外植体,采用3种不同的消毒方法,通过统计污染率、褐化死亡率和成活率,比较变异冬红果茎段较适宜的消毒方法。[结果]用0.05%氯化汞消毒的最佳方式为氯化汞消毒20 min,污染率为17.78%,褐化死亡率为11.11%,成活率为71.11%;用0.1%氯化汞消毒的最佳方式为氯化汞消毒8 min,污染率为22.22%,褐化死亡率为20.00%,成活率为50.00%;用75%乙醇结合0.1%氯化汞消毒的最佳方式为75%乙醇30 s结合0.1%氯化汞10 min进行消毒,污染率为5.56%,褐化死亡率为55.56%,成活率为38.89%。[结论]变异冬红果茎段最佳的消毒方式为0.05%氯化汞消毒20 min,污染率为17.78%,褐化死亡率为11.11%,成活率为71.11%。
[Objective]To establish the sterile system for the first generation of mutant Malus spectabilis.[Method]With mutant Malus spectabilis stem section as explant,three disinfection methods were adopted.By comparing the pollution rate,the mortality of browning and the survival rate,the suitable method of disinfection was selected.[Result]The optimal disinfection method with 0.05%mercuric chloride solution was that the mercuric chloride solution was sterilized for 20 min,the contamination rate was 17.78%,the mortality rate of browning was 11.11%,the survival rate was 71.11%.The optimal disinfection method for mercury solution was disinfection of mercuric chloride solution for 8 min,the contamination rate was 22.22%,browning mortality rate was 20.00%,survival rate was 50.00%.The best disinfection method with 75%alcohol and 0.1%mercuric chloride solution was 75%alcohol for 30 s and 0.1%mercuric chloride solution for 10min,the contamination rate was 5.56%,the browning mortality rate was 55.56%and the survival rate was 38.89%.[Conclusion]The best method was 0.05%mercuric chloride solution disinfection for 20 min,the pollution rate was 17.78%,browning mortality was 11.11%,survival rate was 71.11%.
