伊犁马LTF基因多态性与产奶量相关性分析 被引量:2

摘要 本试验以研究伊犁马乳铁转运蛋白(lactotransferrin,LTF)基因多态性及其与泌乳性状的关系为目标,针对伊犁马LTF基因外显子和启动子设计了17对引物,以伊犁马DNA为模板,通过PCR扩增及PCR-RFLP法检测224匹伊犁马LTF基因多态性,并与其中73匹有产奶量记录的伊犁马进行关联分析,研究该基因多态性与泌乳性状之间的关系。结果显示,在伊犁马LTF基因中检测到1个SNP:c.173G>A,该SNP位点是发生在CDS区上的错义突变,致LTF编码蛋白第58位精氨酸突变为赖氨酸。与其120d日均产奶量进行关联分析,结果显示,虽然该位点不同基因型个体的日均产奶量差异并不显著(P>0.05),但可以发现各基因型的产奶量GG>GA>AA,由此可为中国乳用马的选育提供一定的参考。 This study was to study the relationship between lactotransfferrin(LTF)gene polymorphism and its relationship with lactation.17 pairs of primers were designed for exon and promoter of Yili horse LTF gene.DNA hybrid pool was used as template and PCR product was directly sequenced.PCR-RFLP method was used to detect the expression of LTF(lactotransfferrin)gene polymorphic genotype of 224 Yili horse,and analyzed the relationship between them and lactation traits based on 73 of them.The results showed one SNP was detected in the Yili horse LTF gene:c.173 G>A,which was a missense mutation that occurred in the CDS.It can induce fifty-eighth amino acid of LTF protein mutating from arginine to lysine.Association analysis between SNP site and daily milk yield of 120d was conducted.The results showed that although the milk yield of each genotype was not significantly different(P>0.05),it was found the milk production of each genotype being GG>GA>AA.The results can provide an identifiable ground for the Chinese dairy horse breeding efficiently.
