摘要
以聚丙烯酰肼(PAH)为交联剂,通过席夫碱反应制备了系列氧化透明质酸/氧化羧甲基纤维素/聚丙烯酰肼(oxi-HA/oxi-CMC/PAH)水凝胶,并对水凝胶的溶胀率、降解行为、流变学性能、体外药物释放行为及冻干水凝胶的微观形貌进行了研究。结果表明,oxi-HA6/oxi-CMC6/PAH3水凝胶具有较合适的流变学性能、较高的溶胀率、较好的稳定性和一定的药物缓释能力;药物模型牛血清蛋白(BSA)的体外释放通过扩散机制、溶胀机制和降解机制控制。在37℃pH值7.4的磷酸盐缓冲溶液(PBS)中,oxi-HA6/oxi-CMC6/PAH3水凝胶5 h溶胀率达到16,20 d降解率约为51%,载药水凝胶中BSA的7 d累积释放率约为86%,可应用于药物传输和细胞封装等领域。
A series of oxidized hyaluronic acid/oxidized carboxymethyl cellulose/polyacryloyl hydrazide(oxi-HA/oxi-CMC/PAH)hydrogels were prepared via Schiff base reaction using polyacryloyl hydrazide(PAH)as a crosslinker.The swelling ratio,degradation behavior,rheological properties,in vitro drug release properties of oxi-HA/oxi-CMC/PAH hydrogels and the microtopography of lyophilized hydrogels were investigated.The results showed that oxi-HA6/oxi-CMC6/PAH3 hydrogel exhibited suitable rheology properties,high swelling ratio,good stability and sustainable drug control release capability.The in vitro release of bovine serum albumin(BSA)was controlled through diffusion mechanism,swelling mechanism and degradation mechanism.In phosphate buffered saline(PBS)(pH 7.4)at 37℃,the swelling ratio was about 16 after 5 h,the degradation percentage was about 51%after 20 days,and the cumulative release percentage of BSA was about 86%after 7 days,signifying that oxi-HA6/oxi-CMC6/PAH3 hydrogel could be potentially used in the fields of drug delivery and cell encapsulation.
