改造代谢途径生产芳香族氨基酸衍生物的研究进展

摘要 自然界中许多芳香族氨基酸衍生物具有极高的应用价值,探寻适合工业化生产这些化合物的生物合成途径引起了人们的兴趣。近年来,随着多种生物合成途径不断被发现,代谢工程领域不断发展,在微生物体内重建各种复杂的合成途径变得越来越容易。本文综述了改造微生物代谢途径生产芳香族氨基酸衍生物的最新进展,介绍了白藜芦醇、苄基异喹啉生物碱及类黄酮等芳香族氨基酸衍生物生产的关键成就,讨论了其代谢工程策略的意义及存在的问题。 Many aromatic amino acid-derived products have high applied value.Therefore,interest exists in expressing the biosynthetic pathways of these compounds in organisms that are more suitable for industrial production.Recent years have seen developments in both the discovery of various biosynthetic pathways,as well as development of metabolic engineering tools that allow reconstruction of complex pathways in microorganisms.This review describes the recent advances in modification of metabolic pathways of aromatic amino acid-derived products in various microorganisms.Key achievements in the fermentation of different resveratrol benzylisoquinoline alkaloids,and flavonoids are presented and the significance and the existing problems of the metabolic engineering strategies underlying these accomplishments are discussed.
