摘要
Klotho基因与人类衰老密切相关。Klotho基因敲除小鼠可出现肾脏纤维化等类似于人类衰老的表现。进一步研究发现,Klotho蛋白有抗氧化、抗凋亡、抗衰老、心血管保护和抗纤维化等多种生物学功能。慢性肾脏病(CKD)患者体内Klotho明显下降,说明Klotho在肾脏保护方面发挥重要作用。本文从遗传学角度、Klotho的生理作用及其在CKD中的作用机制方面对Klotho在肾脏保护中的作用进行综述。
Klotho gene is closely related to the aging of human.Klotho knockout mice may appear similar manifestations of human aging,including renal fibrosis.Further studies show that Klotho protein has a variety of biological functions,such as anti-oxidation,anti-apoptosis,anti-aging,cardiovascular protection and anti-fibrosis.The level of Klotho in patients with chronic kidney disease(CKD)decreases markedly,illustrating its important role on protection of kidney.This article reviewed role of Klotho on kidney protection through epigenetics,physiological function,and mechanisms in CKD.
