基质金属蛋白酶2促进胃癌侵袭和浸润

Matrix Metalloproteinases 2 Promotes Invasion and Infiltration in Gastric Cancer

目的比较基质金属蛋白酶2(matrix metalloproteinases2,MMP2)在不同浸润深度胃癌组织中的表达和活性,探讨MMP2与胃癌浸润的关系。方法收集手术切除新鲜胃癌组织46例,其中T1、T2期20例,T3、T4期26例。提取组织总RNA,用RT-PCR方法检测组织中MMP2 m RNA;Western blot方法检测MMP2蛋白;明胶酶谱法检测MMP2酶活性。结果 T3、T4期胃癌组织中MMP2 m RNA相对量为(0.78±0.21),高于T1、T2期(0.54±0.19)(P<0.05);MMP2蛋白相对表达量在T3、T4期胃癌组织中为(0.70±0.14),高于TI、T2期(0.59±0.19)(P<0.05);MMP2酶活性在T3、T4期胃癌组织中也高于T1、T2期(1.44±0.46 vs 1.12±0.53)(P<0.05)。结论胃癌T3、T4期MMP2的m RNA、蛋白表达及酶活性均高于T1、T2期,提示MMP2可能参与胃癌侵袭和浸润。

Objective To compare the expression and activity of matrix metalloproteinases 2(MMP2)in gastric cancer tissues and to analyze the effects of MMP2 on tumor invasion and infiltration.Methods Gastric cancer tissues were collected from 46 patients that underwent surgery,and patients were divided into two groups.T1、T2 group(20 cases)included tumors invaded mucosa or submucosa and did not penetrate serous membrane.T3、T4 group(26 cases)meant tumors penetrated the serous membrane,including infiltrated and not infiltrated adjacent structure.Levels of MMP2 mRNA were detected by RT-PCR.Proteins of MMP2 were examined using western blot.The enzyme activities of MMP2 were evaluated through gelatin zymography.Results Compared with T1、T2 group(0.54±0.19),the expression of MMP2 mRNA was significantly higher in T3、T4 group(0.78±0.21)(P<0.05).The relative protein quantity ofMMP2 was significantlymore abundant in T3、T4 group(0.70±0.14)than that in T1、T2 group(0.59±0.19)(P<0.05).The enzyme activity of MMP2 was significantly stronger in T3、T4 group(1.44±0.46)compared with T1、T2 group(1.12±0.53)(P<0.05).Conclusion MMP2 may promote the invasion and infiltration of gastric cancer.
