摘要
用流式细胞仪对骨髓细胞进行白血病细胞免疫分型,用RT-PCR方法主要检测了急性白血病样品中Hoxa1-13和Meis1基因的表达.结果表明在所有急性白血病患者中CD45和HLA-DR表达阳性,无T系抗原CD3表达,髓系抗原CD33和CD13阳性比例高(76%),B系抗原CD19阳性率为47%,而CD10和CD20阳性率为24%~29%.在急性白血病中Hoxa9和Meis1表达正相关,Hoxa6和Hoxa2表达大多呈现正相关,Hoxa1和Hoxa3在大多数样品中表达负相关.Hoxa3只在CD19阴性CD7阳性的急性白血病样品中表达.结果提示在急性白血病中Hoxa家族基因的表达异常,具有一定的表达相关性,并且免疫表型具有异质性,免疫抗原与Hoxa基因存在着一定的联系.
We performed leukemia immunophenotyping of bone marrow cells with flow c)4ometry,and carried out RT-PCR to detect the expression of Hoxal-13 and Meisl in acute leukemia samples.Results showed that all acute leukemia samples expressed CD45 and HLA-DR,T cell antigen CD3 was not expressed,the positive rate of myeloid antigen CD33 and CD 13 are 76%,B cell antigen CD 19 is 47%while that of CD 10 and CD20 is between 24 and 29%.In acute leukemia Hoxa9 positively correlated with Meisl,in most samples Hoxa6 and Hoxa 13 showed a positive correlation,Hoxal and Hoxa3 are negatively related.Moreover,Hoxa3 was only expressed in acute leukemia samples with CD19"CD7+.These data suggested in acute leukemia Hoxa family genes are abnormally expressed,have certain correlation,and the immunophenotyping is heterogeneous,and immune antigens are closely linked with Hoxa gene expression.
