芥白杂交后代芥菜型油菜遗传物质渗入分析

摘要 为了选育渗入具有芥菜型油菜优良性状的白菜型油菜新资源,拓宽白菜型油菜的遗传基础,改良白菜型油菜的性状,试验将芥菜型油菜(AABB)作为母本,白菜型油菜(AA)作为父本通过杂交获得的F1代,再与父本回交获得BC1F1。并将其连续自交得到BC1F6,从BC1F6代中挑选62个芥白杂交后代单株作为供试材料。利用SSR分子标记技术对母本炉霍油菜的遗传物质在芥白杂交后代中的渗入情况进行分析。结果表明:针对油菜A基因组(A1~A10)设计SSR分子标记引物,并从中筛选得到25对SSR特异性引物,在BC1F6代材料中共扩增出43条带,其中多态性条带占总条带的58.1%;芥白杂交后代中芥菜型油菜遗传物质渗入率为13.95%~32.56%,平均渗入率为20.85%;炉霍油菜A组染色体A1~A10遗传物质都有不同程度的渗入到芥白杂交后代中。通过芥菜型油菜与白菜型油菜进行杂交,能够使种间遗传物质发生交流,从而拓宽白菜型油菜种质资源。 To breed new resources of Brassica rapa with excellent traits and genetic materials of B.juncea,and to broaden the genetic basis of B.rapa and improve its traits,the F1 was obtained by crossing B.juncea(AABB)as the female parent and B.rapa(AA)as the male parent.The BC1F6 was obtained by backcrossing F1 with B.juncea.After continuous selfing,the BC1F6 was obtained,and 62 plants of hybrid offsprings from BC1F6 were chosen as materials.The introgression of genetic material in hybrid offsprings of B.Juncea were analyzed by SSR molecular marker technology.A total of 25 pairs of SSR primers were designed based on the SSR markers of rape A genome(A1-A10).A total of 43 bands were amplified in the B C1F6.Among them,the polymorphic bands accounted for 58.1%.The genetic material infiltration rate of B.juncea in BC1F6 was between 13.95%and 32.56%,with the average infiltration rate of 20.85%.A1-A10 in Agenome of B.juncea has different introgression rates in hybrid offsprings.Crossing B.juncea and B.rapa enable the exchange of interspecific genetic material and broaden the germplasm resources.
