AEG-1与EMT及MRPs相关蛋白在大肠癌组织中表达的临床意义

The clinical significance of the expression of AEG-1,EMT and MRPs related proteins in colorectal cancer tissues

目的:探讨大肠癌组织中星形胶质细胞上调基因-1(astrocyte elevated gene-1,AEG-1)以及上皮间质转化(epithelial-mesenchymal transition,EMT)与多药耐药相关蛋白家族(MRPS)相关蛋白的表达。方法:收集临床标本,采用RT-PCR检测AEG-1的表达,免疫组化(IHC)检测AEG-1、EMT相关蛋白E-cadherin、N-cadherin以及MRPs表达情况,分析三者之间的相关性,进一步分析AEG-1和大肠癌临床病理特征及预后的相关性。结果:免疫组化和RT-PCR结果显示,AEG-1在癌组织中的表达有上调的趋势,并且随着癌症的进展AEG-1的表达增加;EMT相关蛋白E-cadherin、N-cadherin以及多药耐药相关蛋白家族MRPs在癌组织及癌旁组织中的表达,E-cadherin的阳性率为38.1%、N-cadherin的阳性率为52.4%、MRP的阳性率为59.5%;大肠癌组织病理分期及其相关性分析,同癌旁组织比较,AEG-1蛋白在低分化以及Ⅲ-Ⅳ期中表达较高,E-cadherin蛋白在高分化以及Ⅰ-Ⅱ期表达较高,N-cadherin和MRP蛋白在低分化以及Ⅲ-Ⅳ期中表达较高。结论:大肠癌中AEG-1的表达升高,且与EMT及MRPs相关蛋白表达可能存在相关关系。

Objective:To investigate the clinical significance of the expression of AEG-1,EMT and MRPs related proteins in colorectal cancer tissues.Methods:The expression of AEG-1,immunohistochemistry(IHC)for AEG-1,E-cadherin,N-cadherin and MRPs expression in specimens were detected using RT-PCR,the correlation between AEG-1,E-cadherin and N-cadherin was analyzed,and the correlation between AEG-1 and clinical and pathological features of colorectal cancer was further analyzed.Results:IHC and RT-PCR showed that the expression of AEG-1 was up-regulated in cancer tissues,and the expression of AEG-1 increased with the progression of cancer.The positive rate of E-cadherin and N-cadherin was 38.1%and 52.4%,respectively.The positive rate of MRP was 59.5%in cancer tissue with Immunohistochemistry results.Compared with carcinoma adjacent tissue,AEG-1 protein expression was higher in low differentiation and III-IV stages,while E-cadherin protein expression higher in high differentiation and I-II stages.N-cadherin and MRP protein were expressed higher in low differentiation and III-IV stages.Conclusion:The expression of AEG-1 is elevated in colorectal cancer,and is possibly correlated with the expression of EMT and MRPs related proteins.
