摘要
目的:克隆人乳铁蛋白c DNA基因,进而为重组人乳铁蛋白的制备奠定基础。方法:从人静脉血中性粒细胞中提取得总RNA,RT-PCR法扩增人乳铁蛋白c DNA基因后,将其插入到克隆载体p JET1.2,DNA测序法鉴定基因序列。结果:从人静脉血中性粒细胞中分离获得了总RNA,RT-PCR法克隆了人乳铁蛋白c DNA基因,经过DNA测序分析鉴定,其结果与Gene Bank中序列完全一致。结论:成功从人静脉血中性粒细胞中克隆扩增了人乳铁蛋白c DNA基因,为重组人乳铁蛋白的制备奠定基础。
Objective:To clone human lactoferrin cDNA and lay the foundation for the preparation of recombinant human lactoferrin.Methods:Total RNA was extracted from human neutrophils in venous blood.The cDNA of human lactoferrin was amplified by RT-PCR and cloned into the cloning vector pJET1.2.The gene sequence was identified by DNA sequencing.Results:Total RNA was extracted from human neutrophils in venous blood and the cDNA of human lactoferrin was amplified by RT-PCR.The sequence was corresponding with the related sequence in Gene Bank according to DNA sequencing and sequence alignment.Conclusion:Human lactoferrin cDNA is amplified from blood neutrophils in human venous successfully,which lays the foundation for the preparation of recombinant human lactoferrin.
